Background The protein encoded by this gene is a lysosomal acid sphingomyelinase that converts sphingomyelin to ceramide. The encoded protein also has phospholipase C activity. Defects in this gene are a cause of Niemann-Pick disease type A (NPA) and Niemann-Pick disease type B (NPB). Multiple transcript variants encoding different isoforms have been identified. Function Converts sphingomyelin to ceramide. Also has phospholipase C activities toward 1,2-diacylglycerolphosphocholine and 1,2-diacylglycerolphosphoglycerol. Isoform 2 and isoform 3 have lost catalytic activity. [UniProt]

    Flow Cytometry: K562 cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were then incubated in 2% BSA to block non-specific protein-protein interactions and stained with ARG59530 anti-SMPD1 / Acid Sphingomyelinase antibody (right histogram) at 1:25 dilution for 60 min at 37°C, followed by DyLight®488 labelled secondary antibody. Isotype control antibody (left histogram) was Rabbit IgG (1 µg/10^6 cells) used under the same conditions. Acquisition of > 10000 events was performed.
