Ferroptosis is an iron-dependent, regulated form of non-apoptotic cell death driven by the accumulation of lipid peroxides. It shares key features with oxytosis, including depletion of glutathione (GSH), reduced GPX4 activity, and increased lipid peroxidation. Ferroptosis is implicated in diverse physiological and pathological processes, including cancer cell death, neurodegenerative diseases, and ischemia-reperfusion injury. Targeting ferroptosis represents a promising therapeutic strategy for cancer and neurodegenerative disorders.

Ferroptosis is driven by iron overload, lipid peroxidation, and failure of GSH/GPX4 antioxidant system. Lipid metabolism contributes by supplying polyunsaturated fatty acids (PUFAs), which are incorporated into membrane phospholipids and serve as substrates for peroxidation. Iron metabolism increases intracellular iron levels, facilitating Fenton reactions that generate ROS and promote lipid peroxidation. System Xc⁻-GSH-GPX4 axis functions as the primary antioxidant defense, preventing lipid peroxidation and protecting against ferroptosis. Collectively, the core regulatory mechanisms of ferroptosis involve iron metabolism, lipid metabolism, and System Xc⁻-GSH-GPX4 pathway.
