    Immunofluorescence: 293 cells transfected with N-terminal GST tagged gene. Cells were fixed with 4% paraformaldehyde for 30 min at RT, permeabilized with 0.1% Triton X-100 for 10 min at RT then blocked with 10% goat serum for 30 min at RT. Cells were stained with ARG66338 anti-GST tag antibody [SQab1886] (red) at 1:40,000 and 4°C. DAPI (blue) was used as the nuclear counter stain.

    Flow Cytometry: 293 cells transfected with C-terminal (red) and N-terminal (blue) GST tagged gene. The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% TritonX-100 for 15 min. The cells were stained with ARG66338 anti-GST tag antibody [SQab1886] at 1:2,000 dilution in 1x PBS/1% BSA for 30 min at RT, followed by Alexa Fluor® 488 labelled secondary antibody. Unlabelled sample (black) was used as a control. .

    Western blot: 1) 2 µg of 293 cell lysates transfected with N-terminal GST tagged gene, 2) 2 µg of 293 cell lysates transfected with C-terminal GST tagged gene, 3) 10 µg of Mock 293 cell lysates stained with ARG66338 anti-GST tag antibody [SQab1886] at 1:10000 dilution, and 4) 20 ng of two fusion proteins, one (45 kDa) with GST tag on C-terminal, the other (83 kDa) with GST tag on N-terminal stained with ARG66338 anti-GST tag antibody [SQab1886] at 1:2000 dilution.

    Immunoprecipitation: 0.2 mg of 293 whole cell lysates transfected with C-terminal GST tagged gene immunoprecipitated (1:50) and stained with ARG66338 anti-GST tag antibody [SQab1886]. 1) ARG66338 IP in 293 whole cell lysates transfected with C-terminal GST tagged gene, 2) PBS instead of ARG66338 in 293 whole cell lysates transfected with C-terminal GST tagged gene, and 3) 293 whole cell lysate transfected with C-terminal GST tagged gene, 2 µg (input).
