Cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells utilize perforin / granzyme B pathway for killing infected cells or tumor cells. Granzymes are proteases that are packaged in cytotoxic granules with perforin. Perforin is a membrane-disrupting protein that facilitates the delivery of granzymes into the target cell. The granzymes initiate caspase-dependent or -independent cell death.
