Examining Th1/ Th2/ Th17/ Treg cytokines by Multiplex ELISA Kit

CD4+ T cells play a critical role in mediating immune responses to a variety of pathogens as well as in tumor immunity. They are functionally divided into several subsets such as Th1, Th2, Th17, and Treg. The four subsets are antagonistic to each other. The imbalance of Th1/Th2/Th17/Treg is associated with the pathogenesis of autoimmune diseases and oncogenesis.

Th1 cells are induced by IL-12 which promotes STAT4 activation that subsequently induces transcription factor T-bet to activate Th1 cell differentiation. Th1 cells produce IL-2 and IFN-γ and are involved in M1 macrophage polarization and cellular immunity.

Th2 cells are induced by IL-4 which promotes STAT6 activation that induces transcription factor GATA3 to activate Th2 cell differentiation. Th2 cells produce IL-4, IL-5 and IL-13 and are involved in humoral immunity and anti-parasitic infections.

Th17 cells are induced by TGF-β and IL-6 which promotes STAT3 activation that induces transcription factor RoRγt to activate Th17 cell differentiation. Th17 cells produce IL-17 and IL-22 and are involved in the host defense against bacteria and fungi by inducing neutrophil chemotaxis.

Treg cells are induced by TGF-β and IL-2 which promotes STAT5 and transcription factor FoxP3 to activate Treg cell differentiation. Treg cells produce IL-10 and TGF-β and are involved in immune tolerance.
