“Satiety hormone” Leptin links obesity to high blood pressure

Leptin is made by fat and circulates in the bloodstream to reach the brain, where it acts as a signal for energy reserves, adjusting both energy expenditure and the sensation of hunger. Hence, it is sometimes referred to as the “satiety hormone”. Leptin also drives the increase in blood pressure that occurs with weight gain process.

Being obese or overweight is a major risk factor for the development of high blood pressure and cardiovascular disease. A number of factors may be involved, but the exact explanation for the link between these two conditions has been unclear.

Published in December 2014 the journal Cell, Michael Cowley and his research team showed that mice with normal Leptin signaling developed high blood pressure when they became obese on a high fat diet. These effects were not seen in mice that lacked Leptin or where Leptin was unable to work because of a defect or blockade on its receptor. Furthermore, human with loss-of-function mutations in Leptin and the LepR have low blood pressure despite severe obesity. These experiments demonstrate that Leptin signaling is necessary for obesity-induced high blood pressure. The researchers are now investigating the precise pathways in the brain by which Leptin acts to regulate blood pressure, and they believe that these findings could offer help to fight obesity and associated problems such as high blood pressure and cardiovascular diseases. According the previous research, Leptin signaling induces Jak2, PI3K, Erk1/2, and Stat3 activation. arigo provides many useful Antibody Duos/Panels to investigate the downstream pathway of Leptin signal transduction. The Phospho-Jak2 Antibody Duo (ARG30159), Phospho-Stat3 Antibody Duos (ARG30027), Phospho-Erk1/2 Antibody Duo (ARG30026) and Jak2/Stat3 pathway Antibody Panel (ARG30158) are helpful tools for the study of the Leptin downstream signaling. Otherwise, the Leptin ELISA Kit (ARG80126) could be used to detect the changes in Leptin level in blood samples.
