Cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells are effector lymphocytes that utilize Perforin/Granzyme B-induced apoptosis pathways to defend against virally-infected or transformed tumor cells. Cytolytic granules Perforin (PFN) and a family of serine protease including Granzyme B are secreted via exocytosis and induce apoptosis in target cells through the activation of caspases. Perforin deficient mice exhibit reduced ability to clear many viral and intracellular pathogens and are highly susceptible to B-cell lymphoma, tumors and metastases.
