A new role of fat – factory of circulating miRNAs
Adipose tissue is an endocrine organ that secretes a number of proteins, termed adipokines, which play important roles in fat metabolism, feeding behavior, hemostasis, vascular tone, energy balance, and insulin sensitivity. Adipokines raise a great interest as the association of therapeutic possibility for type 2 diabetes, appetite, obesity, and atherosclerosis.
A new role for adipose tissue was reported in Nature recently. Besides releasing adipokines, adipose tissue is also a major source of circulating exosomal miRNAs which are associated with a variety of diseases, including cancer, diabetes, obesity, and cardiovascular disease.
Thomou et al., showed that mice with an adipose-specific knockout of Dicer, the miRNA-processing enzyme, exhibit a decreased levels of circulating miRNAs. The decrease of circulating miRNAs in KO mice can be restored by transplantation of normal adipose tissue. The authors further demonstrated that fat-derived exosomal miRNAs regulate hepatic FGF21 and liver gene expression.
This is the first article reported that adipose tissue is an important source of circulating miRNAs which have far-reaching systemic effects on the regulation of mRNA expression and translation.
