| Background | SOX10 is a member of the SOX (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. The encoded protein may act as a transcriptional activator after forming a protein complex with other proteins. This protein acts as a nucleocytoplasmic shuttle protein and is important for neural crest and peripheral nervous system development. | Function | SOX10 is a transcription factor that plays a central role in developing and mature glia. Specifically activates expression of myelin genes, during oligodendrocyte (OL) maturation, such as DUSP15 and MYRF, thereby playing a central role in oligodendrocyte maturation and CNS myelination. Immunofluorescence: A375 cells were fixed with 4% paraformaldehyde for 30 min at RT, permeabilized with 0.1% Triton X-100 for 10 min at RT then blocked with 10% goat serum for 30 min at room temperature. Cells were stained with ARG66245 anti-SOX10 antibody [SQab1734] at 1:10000 and 4°C. DAPI (blue) was used as the nuclear counter stain.

Flow Cytometry: A375 cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% TritonX-100 for 15 min. Cells were then stained with ARG66245 anti-SOX10 antibody [SQab1734] (blue) at 1:1000 dilution in 1x PBS/1% BSA for 30 min at room temperture, followed by Alexa Fluor® 488 labelled secondary antibody. Unlabelled sample (red) was used as a control.
