Application Note IHC-P: Antigen Retrieval: Boil tissue section in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at RT for 20 min.

Background Cyclin D1 or PRAD-1 or bcl-1 is one of the key cell cycle regulators, and functions in association with cdk4 and / or cdk6 by phosphorylating the Rb protein. It is a putative proto-oncogene overexpressed in a wide variety of human neoplasms including mantle cell lymphomas (MCL).

PTM Phosphorylation at Thr-286 by MAP kinases is required for ubiquitination and degradation following DNA damage. It probably plays an essential role for recognition by the FBXO31 component of SCF (SKP1-cullin-F-box) protein ligase complex.
Ubiquitinated, primarily as 'Lys-48'-linked polyubiquitination. Ubiquitinated by a SCF (SKP1-CUL1-F-box protein) ubiquitin-protein ligase complex containing FBXO4 and CRYAB. Following DNA damage it is ubiquitinated by some SCF (SKP1-cullin-F-box) protein ligase complex containing FBXO31. SCF-type ubiquitination is dependent on Thr-286 phosphorylation (By similarity). Ubiquitinated also by UHRF2 apparently in a phosphorylation-independent manner. Ubiquitination leads to its degradation and G1 arrest. Deubiquitinated by USP2; leading to its stabilization.

    Western blot: 30 µg of HeLa, SW480, 3T3, and Mouse kidney lysates stained with ARG52923 anti-Cyclin D1 antibody [SP4] at 1:500 dilution.

    Immunofluorescence: H1793 cells were fixed with cold methanol:acetone (1:1) for 5 minutes at -20°C, permeabilized with 0.3% Triton X-100 for 30 min at RT then blocked with 1% goat serum for 30 min at RT. Cells were stained with ARG52923 anti-Cyclin D1 antibody [SP4] (red) at 1:100 dilution for 1 hour at RT and DAPI (blue) was used as the nuclear counter stain.
